Culture media

Seawater-based
Medium name Description

20% Z8 SW MEDIUM

200 ml of 100% Z8 diluted in 800 ml of GF/C filtered seawater ( ca 32-34 PSU), autoclaved.

ES

L1

L1 (L1-Si, L1-Si+NH4)

The L1-medium is a modification of the f/2 medium. The difference is a broader trace metal composition in L1. We use it as standard medium for marine diatoms. The L1-trace metal solution is used also in many other media.

Modified Johnsons J/1

This medium is used for Dunaliella salina.

MV

We have used MV (Marine medium with Vitamins) for many years as a standard medium for marine benthic algae. This is the medium named ESI by Tatewaki (1966) but supplemented with vitamins.

TL

This medium is a modification of an Erd-Schreiber medium.

Freshwater-based
Medium name Description

AF-6 Medium (modified*)

DY-V Medium

L16, L16-V

This medium was originally formulated for growth of Peridinium gatunense from Lake Kinneret (Lindström 1991), but works very well for other dinoflagellates as well.

MWC/MWC+Se

We use the recipe in Guillard et al. (1972), but often use a slight modification according to Rengefors (pers. com.): TES- is used instead of TRIS buffer and selenium is added.

NF2/NF2A

This medium is a slight modification of the Bs medium (Christensen 1982). Bs is a modification of the classic freshwater media of Bold and Bristol.

O

This medium (O stands for organic nutrient mixture) is useful for bacteria eating algae like Ochromonas viridis.

SC/SP

Spirulina Medium

Z8

This is our standard medium for blue-green algae.
